Cortical damage or a spatial discrimination control task with similar motor and motivational demands but no dependence on prefrontal cortex. The effects of guanfacine on performance of the delayed alternation task were assessed in additional groups of wild-type versus 2A-AR mutant mice. We observed that functional loss of the 2A-AR subtype, unlike knock-out of the 2C-AR subtype, weakened performance of the prefrontal cortical task without affecting learning and resulted in loss of the beneficial response to guanfacine. These data demonstrate the importance of 2A-AR subtype stimulation for the cognitive functions of the prefrontal cortex and identify the molecular substrate for guanfacine and novel therapeutic interventions. Three 2-AR subtypes have been cloned in humans: the 2AAR, 2B-AR, and 2C-AR, with the 2A-AR and 2C-AR, but not the 2B-AR, subtypes localized in the PFC for review, see Aoki et al., 1994; MacDonald et al., 1997 ; . Previous studies in monkeys used agonists with varying affinities for the 2A-AR, 2B-AR, and 2C-AR to attempt to dissect the subtype or subtypes underlying the cognitive-enhancing, hypotensive, and sedating actions of these agents Arnsten et al., 1988; Arnsten and Leslie, 1991 ; . These pharmacological analyses indicated that the cognitiveenhancing effects were mediated by the 2A-AR, whereas the hypotensive and sedating actions were mediated by the 2B-AR and 2C-AR. It is now known that these conclusions were erroneous. Studies of mice with genetically altered 2-AR subtypes demonstrated conclusively that the 2A-AR subtype mediates the hypotensive actions of 2-AR agonists Link et al., 1996; MacMillan et al., 1996 ; , and the sedative effects of these agents involve 2A-AR actions as well for review, see MacDonald et al., 1997 ; . The faulty conclusions from the previous pharmacological studies most likely arose from several factors: 1 ; there are no highly selective 2A-AR, 2B-AR, or 2C-AR agonists or antagonists to dissociate actions between the subtypes; 2 ; many.

Joint capsule, tendinous attachment were sutures back to the corresponding sites. Results: During the follow-up period two patients expired from metastatic disease. Follow-up lost in two patients. In long term follow-up all patients regained stable painless limited range of motion. Based on the MSTS functional results patients had excellent results with respect to pain control, emotional acceptance and manual dexterity. And poor with active motion. Discussion: Reconstructive modality proximal humerus following wide resection is variable. Extent of surgical defect, status of abductor mechanism, patient's age, level of activity, expectation, and long-term prognosis should be taken into consideration. Reconstruction using osteoarticular allograft obtained good early result. However, the long-term result is associated with a high complication rate related to subchondral fragmentation, collapse, leading to multiple revision procedures. In selected patients, these complications can be overcome by the use of the allograft prosthesis composite, which has proven to result in durable satisfactory outcome, well accepted by the patients and halcion.

1. Experiments were performed to determine the role of intra- and extracellular a Ca2 + during contractions of canine saphenous veins evoked with 'x '- and a2 '-adrenergic agonists. 2. After incubation in Ca2 + -free medium for 45 min noradrena~lihe, clonidine and phenylephrine caused contractions, reaching a maximal amplitude rapidly and then declining to a lower stable level within 6 min. The maximal amplitude amounted to about 10 % of the response to the agonists in physiological saline containing Ca2 + ions. After a previous exposure to one of the a-adrenergic agonists in Ca2 + -free medium, no peak response was observed when the preparations were exposed to any of the agonists a second time. The stable smaller contractions which followed the peak response could however be repeated after a previous exposure to one of the agonists. 3. In normal physiological salt solution PSS ; , iproveratril depressed the maximal response to all three agonists to about 60 % of the control value. However, the shift of the median effective dose ED50 ; to the right was larger for clonidine than for the other two agonists. 4. Noradrenaline, clonidine and phenylephrine all at 10-5 M ; increased the uptake of 45Ca. Iproveratril reduced this increased uptake of 45Ca. 5. Noradrenaline, clonidine, guanfacine and phenylephrine augmented the efflux rate of 45Ca. After exposure to one of the agonists, noradrenaline caused no or only a moderate increase of the efflux rate of 45Ca. 6. The policy conundrum faced by every national government is how to ensure that it retains the capacity to govern while also providing its citizens with the benefits of a more globally integrated economy. How do governments pursue national economic in terests in a world where the principal organizer of economic activity, the firm, is more likely to have global rather than national interests? How can trade agreements be harnessed to provide the interface between national governance and global commerce? How can judicial oversight of governance be implemented more fully at the international level. 73 To date, governments have been prepared to address some of the issues raised by deepening global integration as trade-related issues, such as in the WTO Agreement on Trade-Related Investment Measures TRIMs ; . Environmental groups are similarly calling for negotiations about trade-related environmental measures, labour unions are contemplating trade-related fair labour standards, and human rights groups are considering the scope for using trade agreements to strengthen compliance with international human rights conventions. Without commenting on the merits of each of these issues, the extent of demands on the trading system bears testimony to the high level of flux in thinking about appropriate rules to govern a global economy.
