Nother new large study of weight and mortality confirms that the lowest mortality appears to be at moderate body weights at all ages for both men and women, but findings by race are labelled "enigmatic" in an accompanying editorial.1 Results come from the Cancer Prevention Study II, a prospective US study that followed over a million people from 19821996. They are reported by epidemiologist Eugenia E. Calle, PhD, and her colleagues at the American Cancer Society in the October 1999 issue of the New England Journal of Medicine.2 The results took into account many variables, including age average age at the beginning of the study was 57 ; , education but not income ; , physical activity, alcohol use, marital status, use of estrogen supplements in women ; , and crude consumption of fats and vegetables. Subjects were divided into four groups according to whether they had ever smoked and whether they reported a history of disease including any current illness or weight loss of 10 lb [4.5 kg] or more in the previous year ; . This subgroup of healthy non-smokers gives a better estimate of the association between body mass index BMI ; and mortality because it has removed the influence of smoking and disease. There were 201, 622 deaths. The association between mortality and high BMI was strongest in healthy people who had never smoked see Figure 1 ; . The heaviest men, presumably with BMIs over 40, had 2.68 times the risk of men with BMIs between 23.5 and 24.9 the reference category ; . The heaviest women had 1.89 times the risk of their reference group. The relationship between mortality and low BMI was strongest for current and former smokers with a history of disease. This increased mortality in lean subjects was explained primarily by cerebrovascular disease, pneumonia, and diseases of the central nervous system. The effects of race were further explored in the healthy non-smoking subgroup. White men with a BMI between 23.5 and 24.9 had the lowest death rate. For white women, the death rate was lowest at a BMI of 22.0 to 23.4. A small but statistically significant increase in the risk of death began at BMIs of 26.5 to 27.9 for men and 25.0 to 26.4 for women. This association appears at lower weights than is reported in other studies, and.
Phosphatidylcholine multilayers, containing 20% water by total sample weight and gramicidin lipid molar ratios up to 1: were aligned by low temperature annealing 600C ; and mechanical stressing. We were able to obtain large 80 , um thick x 40 mm2 area ; monodomain defect-free multilayers containing -10'7 uniformly oriented gramicidin channels. The alignment of lipid multilayers was monitored by conoscopy and polarized microscopy. The smectic defects, which appeared during the alignment process, were identified and dissolved. The incorporation of gramicidin into the multilayers in the form of transmembrane channels was indicated by its circular dichroic CD ; spectrum. A well-defined CD spectrum of uniformly oriented gramicidin channels was obtained. The oriented samples will allow spectroscopic studies of the ion channel in its conducting state and diffraction studies of the channel-channel organization in the membrane.
The approach differs significantly from that taken by others. The influence of polarization of the polar groups by the ion is treated explicitly. This has the advantage that the charge distribution of groups near an ion can be perturbed by the ion; their properties can and do ; differ substantially from the group average. There is, however, a price. The model parameters are less well optimized than those used in more standard force fields GROMOS, AMBER, CHARMM ; . Furthermore, the basic units are dipolar groups CO and NH ; , not charged atoms C, 0, N, and H ; . Consequently orientational alignments due to steric influences may not be handled as well. Additionally, the charges in the polar groups are not separated, but treated as point dipoles. However, as the mean helix structure in the thermalized system does not differ greatly from that of the 0 K configuration, it would seem that the model incorporates the essential features of the gramicidin helix. Phosphorylation of villin increased its actin-depolymerizing property. Fluorescence measurements show a 22% n 9, p 0.01 ; increase in actin severing by VILT WT compared with VIL WT. These results are consistent with an increase in the actin depolymerizing function of phosphorylated villin. These data support our hypothesis that tyrosine phosphorylation of villin and a decrease in the intestinal cell F-actin content are not mutually exclusive. Tyrosine Phosphorylation Alters the Actin Filament Organization--To further explore the correlation between phosphorylation and regulation of the actin-modulating properties of villin, we examined the effects of VIL WT and VILT WT on actin filament morphology. Actin polymerized in the absence of villin shows long, curved F-actin filaments that are not cross-linked Fig. 3A ; . In the presence of saturating amounts of VIL WT relative to actin villin: actin, 1: 2 ; and in the absence of Ca2 , VIL WT organizes the F-actin filaments into long, straight, closely aligned, well organized, tight bundles Fig. 3, B and B ; . In contrast, phosphorylation of villin led to reorganization of the actin filaments, which now showed thicker bundles, which at higher magnification appeared to have loose, poorly organized filament structure. The bundles showed no distinct alignment of filament but contained loosely packed filaments with some associated electron dense material Fig. 3C ; . There was also an appreciable decrease in the average length of the filaments Fig. 3C ; . Thus, although VILT WT can bind actin filaments albeit with lower binding affinity for actin, it does not bundle filaments like VIL WT. Negative charges exposed on the surface of the actin filaments favor their alignment into bundles, and polyanions can disintegrate actin bundles into single filaments 14 ; .
